The truck startup and runs sometimes after driving it 5 miles and turning off the engine. When you go restart the engine it won't kick over, even if you spray starter fuel in the throttle body inlet. I need help?

Never use ether on a diesel engine you are not going to yank out and rebuild. In other words if it doesn't start fix the damn problem before making it worse and extremely expensive! The only two times I have used it myself both engines we were going to yank anyway. One already had a hole in a cracked piston. The other is a current project with a shot timing set and no rings left.
Duramax engines don't have a lift pump so any air leaks will give you starting problems. Try using the hand primer to prime the fuel system. Some places to check for air leaks are the fuel filter: if it is a plastic Wix this is your problem as they leak at the WIF threads. Next is a primer seal leak. Then cracked fuel filter housing.
Cranking RPM is critical to start a diesel. Make sure both batteries are good by disconnecting them and testing each. If the engine sounds like it is cranking slow: IT IS! And of course won't start.
Further diagnosis will require a scanner to watch rail pressure and check sensors for cranking RPM etc.
