Dad recently obtained a 2000 GMC C3500 C&C 5.7L/4L80E. I was wondering, can the TCC lockup switch mod work on a gasser?

No reason why it wouldn't... you're putting the mod on the tranny, not the engine.
Figuring out which wire it is could be a little tricky... I dunno what the pinouts are on the PCM on the gasser. I do know that line 'S' out of the tranny should still be the right one... head down to a transmission shop and ask 'em.
